Modeling Enzymes Inhibition During Microparticles Formation

چکیده

Abstract. A mathematical model of enzymes inhibition during microparticles formation has been developed. The model is based on radial substrate diffusion through diffusion layer that surrounds microparticle. The inhibition of enzymes was explained by enzyme adsorption on surface of the microparticles and substrate diffusion limitation through the diffusion layer. The diffusion module is dependent on dimensions of the microparticle. The microparticles with diameter 2μm may ensure 50% of an apparent activity decrease if half of peroxidase has been adsorbed. Further adsorption of peroxidase and aggregation of the microparticles increases the apparent inhibition.

Extravasating Neutrophil-derived Microparticles Preserve Vascular Barrier Function in Inflamed Tissue

Emerging evidence suggests that gap formation and opening of the endothelial junctions during leukocyte extravasation is actively controlled to maintain the integrity of the vascular barrier. While the role for endothelial cells to this process has been well defined, it is not clear whether leukocytes are also actively contributing to endothelial barrier function. We have recently showed that e...

Megakaryocyte-derived microparticles: direct visualization and distinction from platelet-derived microparticles.

Platelet microparticles are a normal constituent of circulating blood. Several studies have demonstrated positive correlations between thrombotic states and platelet microparticle levels. Yet little is known about the processes by which platelet microparticles are generated in vivo. We now characterize microparticles derived directly from megakaryocytes. Video microscopy of live mouse megakaryo...

Endothelial-derived microparticles: Biological conveyors at the crossroad of inflammation, thrombosis and angiogenesis.

Endothelial microparticles (EMP) are complex vesicular structures that can be shed by activated or apoptotic endothelial cells. EMP are composed of a phospholipid bilayer that exposes transmembrane proteins and receptors and encloses cytosolic components such as enzymes, transcription factors and mRNA derived from their parent cells. Thus, EMP behave as biological conveyors playing a key role i...

Microparticle generation and leucocyte death in Shiga toxin-mediated HUS.

BACKGROUND Shiga toxin-induced haemolytic uraemic syndrome (STEC-HUS) is an acute multisystem disorder characterized by renal failure, neurological dysfunction, haemolysis and intravascular thrombosis. Circulating microparticles originating from a number of cell types including thrombocytes and leucocytes are elevated in paediatric patients. In vitro data also suggest modification of leucocyte ...

Study of the immune adjuvant effect of chitin microparticles on plasma levels of IgG1 and IgG2a during immunization against Leishmania infection in BALB/c mice

Background: There are reports on the ability of chitin microparticles to modulate the TH1 and TH2 responses, depending on the size and administration route. The purpose of the present study was to investigate the immunoadjuvant effects of the small-sized (less than 40 microns) chitin microparticles (CMP) in vaccination against Leishmania major for preventing leishmaniasis in BALB /c mice by ...
